Bioactive compounds from natura sources - Chemical characterization and biological properties

The analysis of nutrients will include determination of proteins, fat, ash and carbohydratesm as also individual profiles in sugars (high performance liquid chromatography coupled to refraction index detector, HPLC-RI) and fatty acids (gas chromatography coupled to flame ionization detector, GC-FID). The analysis of nutraceuticals will include determination of phytochemicals such as tocopherols (HPLC-fluorescence), phenolic compounds (HPLCcoupled to a diode array and mass spectometry, HPLC-DAD-MS) organic acids and ascorbic acid (ultrafast liquid chromatography coupled to photodiodearray detector (UFLC-PDA) and steroids (HPLC-UV). Biological characterization will be focused on bioactive properties such as antioxidan activity, measured by chemical (free radical scavenging activity and reducing power) and biochemical assays with animal cells (brain cells homogenates; lipid peroxidation inhibition), and of antitumor activity using human tumor cells lines by sulphorhodamine assay (MCF-7-breats cancer, NCI-H460-non-small cell lung cancer, HCT-15-colon cancer, AGS-gastric cancer and HepG2-hepatocellular carcinoma). The toxicity in non-tumour cells (animal hepatocyctes) will be also tested.
